General procedure: Fe 3 O 4 /COSbeta-CD-SO 3 H NPs (V) (0.02 g) were added to a mixture of isatin (1 mmol, 0.147 g), 5,5-dimethylcyclohexane-1,3-dione (dimedone) (1 mmol, 0.140g), and malononitrile (1mmol, 0.066g) in water (4mL). The mixture was magnetically stirred at 50C, for 20min. Reaction progress was monitored by TLC (n-hexane:EtOAc, 3:1). Upon reaction completion, the catalyst was removed by an external magnet, washed with water (10mL), and dried overnight to be ready for the next run. The formed solid was filtered off, washed with warm water, then dried to obtain pure products (0.328g, 98%).

General procedure: The preparation was divided into two steps. Firstly, substituent indoline-2,3-dione (1 equiv) was dissolved in ethanol (10 mL), hydrazine hydrate (2 mL) was added. The reaction mixture was refluxed with magnetic stirring for 3 h and cooled to 0 C, then sodium hydroxide (3 equiv) was added, refluxed for 0.5 h and cooled to room temperature, then 150 mL water was added. The mixture was acidified by adding 2 N hydrochloric acid to pH 2, and extracted twice with dichloromethane. The organic filtrate was washed twice with brine, dried (Na2SO4), and then concentrated to give corresponding indolin-2-one by high-vacuum drying. Secondly, a mixture of the corresponding indolin-2-one (1 equiv) and furan-2-carbaldehyde (1 equiv) in ethanol was added two drops of piperidine and refluxed with magnetic stirring for 5 h. After the mixture cooled, the precipitate was filtered, washed with cold ethanol, dried, and recrystallized from methanol to afford the terminal product.

(0331) A mixture of potassium phthalimide (100.0 g, 539.9 mmol, 1.0 equiv.), potassium carbonate (11.2 g, 81.0 mmol, 0.15 equiv.) and N,N-dimethylformamide (DMF) (200 mL) is treated with chloroacetone (47.3 mL, 54.9 g, 593.9 mmol, 1.1 equiv.) at a rate to control the internal temperature at not more than 40 C. After the addition is complete, the reaction mixture is stirred for about 12 hours at about 20-25 C. Water (333 mL) is charged to the reaction mixture at a rate to control the internal temperature at not more than 30 C. The mixture is stirred at about 20-25 C. for about 1 hour, and then the mixture is filtered. The filter cake is washed with water (200 mL), and the product is dried at about 70-80 C. under vacuum with a nitrogen flow for about 12 hours. The product phthalimidoacetone (VII-j) is obtained as a white solid (90.2 g, 97.3 wt. % purity, 80% yield).

After extraction with DCM, the organic layer was washed with brine, dried over anhy. Na2S04, filtered and concentrated in vacuo to give a crude product containing a mixture of two regioisomers (5.82 g, 100%). MS: 212.0 (M+H+).

To a 100mL three-necked flask fitted with a stirring blade, a thermometer, acondenser, indoline derivative tartrate salt (purity99.9%) 5.0 g, butyl acetate 15 mL and separately adjusted 20% aqueous potassiumcarbonate solution 14mL were added & stirred, and warmed in the vicinity of45 & stirred, and it was made sure that the solid was dissolved, to obtain aprimary reaction solution.To the primary reaction liquid obtained in theneutralization step, 1- (2-methanesulfonyloxy-ethoxy) -2-(2,2,2-trifluoroethoxy) benzene 3.67g was added as N- alkylating agent and stirred,after stirring for 18 hours under heating to reflux, then cooled to about 30 C, to obtain a secondary reaction solution (reaction conversion rate 95.4%).The aqueous layer was removed from the secondary reaction mixture, the resulting organic layer was washed successively with saturated aqueous sodiumbicarbonate solution 10 mL and 10% aqueous sodium chloride 10 mL, to obtain butylacetate solution (purity 81.0 %, dialkyl body content 9.4%) of N-alkylated indoline derivative.

A mixture of 6-bromoisoindolin-1-one (1.0 g, 4.72 mmol), Pd(dppf)Cl2 (104 mg, 0.14 mmol), K2CO3 (1.96 g, 14.2 mmol) and 3-pyridylboronic acid (580 mg, 4.72 mmol) in dioxane (45 mL) and water (5 mL) was heated to 100 C. overnight. After cooling, the mixture was poured into water and extracted with EtOAc (3×100 mL). The combined organic fractions were washed with brine and dried over sodium sulfate. The solvent was evaporated and the residue was purified by column chromatography on silica gel (1%-50% EtOAc in pet. ether) to afford 85B (500 mg, 50% yield) as an off-white solid: 1H NMR (400 MHz, DMSO-d6) delta 8.94 (d, J=2 Hz, 1H), 8.67 (s, 1H), 8.60 (dd, J=4.8 Hz, 1.2 Hz, 1H), 8.16-8.13 (m, 1H), 7.96-7.94 (m, 2H), 7.71 (d, J=8.8 Hz, 1H), 7.52-7.49 (m, 1H), 4.44 (s, 2H); ESI m/z 211.0 [M+H]+.
